Transferred proteins (>200 ng/band) appear as red bands on a pink background. Ponceau S staining is simple, rapid, and reversible, which is especially advantageous when the blot needs to be reused for a second detection method (e.g., immunoblotting). Ponceau S is a water-soluble anionic diazo dye, with the molecular formula C22H12N4Na4O13S4 and molecular weight of 760.6. It carries a negative charge and can bind to positively charged amino acid residues.
