The fundamental principle of staining lies in the fact that nucleic acids within the cell nucleus are negatively charged and acidic, making them prone to binding with the positively charged, alkaline hematoxylin dye, thus becoming stained. With a hematoxylin concentration of 0.5%, the stained nuclei appear vividly blue or bluish-purple.
